光合成微生物Synechocystis sp. PCC 6803中心代謝酵素の定量プロテオミクス

A method for quantitative proteome analysis of the central metabolism-related enzymes in photosynthetic microorganism, Synechocystis sp. PCC 6803 (Syn6803), was developed by ultra-fast mass spectrometry. In order to find suitable SRM transitions for sensitive analyses, several well-behaved peptides were selected from the possibly digested fragments of a target protein. Suitable SRM transitions were selected from an analysis of peptide samples prepared from Syn6803 by a method including SRM transitions (up to 500 transitions) of all y- and b-series fragment ions generated from the [M+2H]2+ of peptides. A comparison of the enzyme levels in the Syn6803 cells demonstrated that expression of several enzymes was significantly up-regulated in the autotrophic condition for an active carbon fixation.
