LC/MSへの応用に向けた大気圧赤外レーザー脱離イオン化法の開発

As the new ionization method for a rapid analysis of biomolecules, atmospheric pressure infrared laser desorption/ionization has been researched. Since liquid samples are directly ionized by the irradiation of infrared laser at atmospheric pressure without any additional matrices, this ionization has been expected as the interface of liquid chromatography/mass spectrometry (LC/MS). In this study, the ion source of ESI-MS was modified to an atmospheric pressure laser ion source. By using a porous stainless steel plate “frit”, this ionization method is applicable to the analysis of continuous-flow liquid samples. The ion source was directly connected to the high-performance liquid chromatography (HPLC) system, and online LC/MS analysis of a mixture of two standard samples, angiotensin II and reserpine, was performed. As a result, two samples from HPLC was ionized and detected at MS separately. In addition, the chromatogram was stable during the measurement in 50 minutes. It was confirmed that atmospheric pressure infrared laser ion source using a frit is applicable to LC/MS. This ionization method can be expected for comprehensive analysis of complex biological samples such as membrane proteins.
