リアルタイム直接質量分析(DART-MS)による角層中アミノ酸の定量法の開発
Analysis of Natural Moisturizing Factors (NMFs) in stratum corneum (SC), such as amino acids, pyrrolidone carboxyric acid and lactic acid, is important in the dermatology research. Conventional analytical techniques for NMFs are liquid chromatography and gas chromatography. However, these techniques are not sufficient in terms of robustness and analysis time resulting from sample preparation including extraction, purification, condensation and derivatization. Direct analysis in real time-mass spectrometry (DART-MS) is a powerful technique in measuring analytes directly without sample preparation and separation. We have developed a rapid, easy and precise analytical technique to quantify amino acids in SC by the combination of DART-MS and stable isotope-labeled internal standard. Good linear correlations were obtained for calibration curves of 12 amino acids and only several minutes were required to determine the mounts of 12 amino acids in one sample.
