In this study, we try to develop the real-time metabolomic analysis system using probe electrospray ionization-mass spectrometry (PESI-MS). To use the PESI-MS as a real-time metabolomic analysis system in vivo, we elongated the ion path through a long tube made of stainless steel. This tube was connected to the original ion inlet by a custom-made connector. This connector has one EVAC port, which is connected to a diaphragm pump to increase the flow rate of the carrier air, and hence assisting the ion transmission over the long separation distance. We termed it “non-proximate PESI-MS” for this system. To evaluate the applicability of non-proximate PESI for real-time metabolomic analysis, we directly analyzed the metabolic profiles of liver tissue in chemically induced hepatitis model mice. The ion signals were strong enough to discriminate the different classes of experimented conditions by the partial least squares-logistic regression (PLS-LR). This results indicate that, non-proximate PESI-MS system allowed us to directly monitor the metabolism and pharmacokinetics in living animals. Additionally, we are now trying to establish software for database construction/statistical analysis. This software strongly supports the data management and screening of significant peaks. Details of this software are also presented.
