血液検体の処理および保存状態が代謝物プロファイルに与える影響

Currently, metabolomics has attracted considerable research interest and applications. However, unlike other omics technology, standardization of analytical method is difficult due to the complexity of the physical and chemical property of metabolites. Therefore, several MS-based analytical platforms are employed in metabolomics.
In the past years, we have developed capillary electrophoresis-mass spectrometry (CE-MS) methods, which is well suited for study of ionic metabolites. CE can provide a high peak resolution for ionic metabolites, while MS provides high selectivity and sensitivity.
In order to acquire high quality metabolomics data, it is important to pay careful attention to sample processing and storage conditions before metabolite extraction. We have quantitatively analyzed the effects of sample collection, treatment, and storage conditions by comparing serum and plasma, and by testing the impact of sample incubation time and temperature, sample storage time and freeze-thaw cycles on CE-MS determined metabolomics results. In this lecture, I would like to explain these results.
