確認イオン比を利用したGAG二糖異性体の一斉定量解析
The glycosaminoglycan (GAG) consists of repeating disaccharides and shows high heterogeneity generated through modifications such as sulfation or epimerization along the backbone. GAG family is classified on the disaccharide unit into four classes, hyaluronan (HA), chondroitin/dermatan sulfate (CS/DS), keratan sulfate (KS), and heparan sulfate/heparin (HS/HP). GAGs are constitutively located on the extracellular matrix of many tissues such as cartilage, skin, and aorta. The structure–function relationships of GAGs and the significance of their alteration in disorders of complex biological systems are indicated.
We tried to simultaneously measure multiple disaccharides derived from mixtures of different GAGs by tandem mass (MS/MS) analysis with specific channels after on liquid chromatography (LC) with a porous graphitized carbon column, Hypercarb. Several isomers having similar retention time and detected with a same channel, were identified with a reference ion ratio. By our method, 23 disaccharides were successfully identified and quantified.
