MSで異性体解析に挑む:シアル酸結合様式特異的誘導体化法 “SALSA” の開発
The goal of this study is to develop a rapid, sensitive and reliable linkage-specific derivatization method for sialic acids. It is more preferable that the derivatization can be applicable not only to glycans, but also to glycopeptides. Toward this goal, a novel derivatization method using alkylamine and condensing reagents was investigated. Amidation using isopropylamine (iPA) resulted in complete discrimination of α2,6- and α2,3-sialic acids by forming iPA-derivatives and lactone forms, respectively. Furthermore, lactone forms could be selectively converted to methylamine (MA) forms by an additional reaction. This Sialic Acid Linkage-Specific Alkylamidation (SALSA) is performed under mild reaction conditions and completed within a relatively a short time.
