超臨界流体クロマトグラフィー質量分析を用いた代謝プロファイリング

A supercritical fluid can be used as the mobile phase in chromatography owing to its favorable properties, such as low viscosity and high diffusivity. Supercritical fluid chromatography (SFC) can provide high-speed and high-resolution separation. Because supercritical carbon dioxide (SCCO2), which is generally used as the mobile phase in SFC, is automatically emitted at room temperature, SFC is most commonly used as a preparative method. However, because of the low polarity of SCCO2, SFC can also be used to perform high-precision biomolecular analysis, especially that of hydrophobic metabolites. The use of a mass spectrometer (MS) with SFC can widen the scope of application of SFC to biological sample analysis. Therefore, we have tried to apply SFC/MS to metabolic profiling. First, we successfully developed lipid metabolomics (lipidomics) system by SFC/MS. Additionally, targeted-profiling methods for various lipids using SFC/MS were also constructed. Furthermore, we developed technologies for combining SFC and supercritical fluid extraction (SFE). We successfully applied SFE/SFC to labile compounds such as CoQ10 and fat-soluble vitamins.
