Differential mobility separation (DMS) is a technique that can separate ions in the gas phase, prior to MS analysis and can complement the LC separation. Ions transmitted through a DMS cell will exhibit 3 types of behavior with respect the compensation voltage (CoV) increasing separation voltage (SV); a-type, b-type and c-type. For a-type, the CoV contuniously increases as SV is raised. For c-type, the CoV continuously decreases (negative values) as SV is raised. And finally, for b-type, as the SV is raised, a decrease in CoV is first observed, followed by an increase in CoV beyond a certain SV voltage (inflection point). Intact antibody and ADC exhibit a c-type behavior in a DMS cell, whereas the light- and heavy-chain will have a-type and b-type behavior, respectively. With 3 different behaviors, this indicates that all 3 species can be separated with DMS cell and consequently, under co-eluting conditions, the DMS cell can separate all form by transmitting them at separate CoV values. Operating with DMS therefore leads to simplified mass spectral information and minimizes risk of convolution. In this present, we show that improved antibody based products can separate with Differential Mobility Separation.
