Mass spectrometry is now one of the most powerful and essential methods for understanding molecular function of proteins. Native mass spectrometry provides exact mass of a protein complex, from which stoichiometry of the interaction can be determined. Using native mass spectrometry, non-covalent nature of the binding between nuclear receptor and organotins were successfully indicated at the first time.
Information on the higher order structure of protein can be obtained by hydrogen/deuterium exchange mass spectrometry (HDX-MS). We have studied on the structure of condensin SMC proteins by X-ray crystallography and HDX-MS. In addition to the identification of sites of binding to DNA, structural alternations upon the interaction were clarified. Impact and importance of mass spectrometry will be introduced with recent examples of studies on protein-protein or protein-low molecular compound interaction.
