LC/MS analysis of oligonucleotides using polymer-based HILIC column having diol group

For development or quality control of oligonucleotide medicines, it is required to analyze them with high sensitivity and high selectivity using LC/MS. Though ion-pair reverse-phase chromatography have been used conventionally, there is a problem that ion-pair reagent tend to remain in LC/MS instrument. In order to solve this issue, a HILIC column Shodex® HILICpak® VN-50 2D with polyvinyl alcohol base packing material modified with diol group was used for the LC. Aqueous solution including ammonium formate / acetonitrile was used as eluent and ESI-MS was used for detection. As a result, it was confirmed to obtain good separation of 2-20mer DNA. This method enables the analysis of oligonucleotides without ion-pair reagent.
