Development of Electrospray-Assisted Laser Desorption/Ionization Source for Imaging of Intracellular Molecules

Imaging of intracellular molecules is important analytical technique to understand dynamics of proteins and metabolites in a cell. In previous study, using mass spectrometry imaging, high spatial resolution (1 μm) was achieved. However, sensitivity of ions is not sufficient. In order to acquire clear and detailed image, it’s necessary to increase amount of ions. In this experiment, to improve ionization efficiency, ELDI (Electrospray-assisted laser desorption/ionization; ELDI) source combining laser desorption ionization (LDI) and electrospray ionization (ESI) under atmospheric pressure and optical system was constructed. Using ELDI, crystal violet (CV) was measured and the effectiveness of ELDI source was evaluated.
