Pharmacokinetic Study of Super-Multicomponent Drug, Such as Traditional Japanese Kampo Medicine, Using High-Resolution Mass Spectrometry Analysis

High resolution (HR)-mass spectrometry analysis enabled us to analyze various compounds present in super-multicomponent products, such as herbal medicines and food stuffs. We used LC-HRMS (Xevo G2-XS quadrupole time-of-flight MS; Waters) to investigate how the active compounds in kampo, a traditional Japanese medicine, contribute to its effects. We performed a pharmacokinetic analysis of kampo ingredients using LC-HRMS analysis.
We set out to semi-quantify the compounds present in the biological samples following the administration of the super-multicomponent drug kampo using surrogate calibrations from other compounds that have the typical skeleton structure of a phytochemical. The use of LC-HRMS methodology allows us to identify the approximate concentrations of potentially active compounds of kampo medicine. For example, we found that many flavonoids exist in the mice plasma after the administration of ninjin’yoeito and the total concentration of the flavonoids is comparatively high.
In conclusion, we found an effective approach for pharmacokinetic analysis of compounds present in super-multicomponent products, like kampo, using LC-HRMS analysis.
