Structure estimation of gram-negative gut bacterial lipid A using untargeted analysis

A method for the analysis of lipid A, an important immune activator of gram-negative bacteria, was developed by using LC-QTOF-MS. The sharp peak of lipid A was observed by employing a metal-free column and high pH basic solvent. MS/MS spectra assignment enabled the structure estimation of lipid A. This method was successfully applied to the analysis of in vitro cultured gram-negative gut bacteria strains. The results suggest the structural diversity of lipid A.
