Wide-range metabolomic analysis of hydrophilic metabolites by next-generation ion chromatography high resolution tandem mass spectrometry

Almost all intermediates of essential metabolic pathways to maintain biological activities are cationic, zwitterionic, or anionic metabolites. Therefore, a profiling for these ionic metabolites provides valuable insight towards understanding metabolic regulation. To date, we have developed methods for the analyses of anionic metabolites and cationic metabolites, respectively, using ion chromatography high resolution tandem mass spectrometer (IC/HRMS/MS). However, in conventional IC system, many of zwitterionic metabolites such as amino acids and their derivatives are removed by a process that uses suppressors to convert the high salt eluent to pure water (i.e., desalting). The aim of this study is to develop an advanced analytical system for wide-range metabolomic analysis of cationic, zwitterionic, and anionic metabolites by IC/HRMS/MS with novel cationic and anionic suppressors.
