Application of MSE in Chiral Metabolomics for Identifying Tissue-Specific Minor Metabolites

Some minor enantiomers of chiral organic acids have enantiospecific roles, thus metabolome analysis including such enantiomers could provide new discoveries. However, simultaneous separation of enantiomers for various compounds is generally difficult. We previously developed a chiral derivatization reagent (S)-1-(4,6-dimethoxy-1,3,5-triazin -2-yl) pyrrolidin-3-amine (DMT-3(S)-Apy), which enables a highly sensitive simultaneous analysis of carboxylic acids, and obtained new insights regarding disease2) or beverages. These analysis suggested that improvement in MS/MS data coverage is required for including low abundant molecules in the chiral metabolomics. In this study, we used DMT-3(S)-Apy and its derivative (D6-DMT-3(S)-Apy) to compare MS/MS data coverage in dependent analysis (DDA) mode and that in the MSE mode. Pooled human serum was used to compare the DDA and the MSE. DMT-3(S)-Apy and D6-DMT-3(S)-Apy were synthesized described previously. Various tissues were collected from C57BL-6J male mice at 8 weeks old. The homogenate of tissues were subjected to the LC/MS analysis. We found that the stomach, cecum, and cerebellum contained various characteristic peaks. Various specific peaks in these tissues were then extracted. We identified some peaks with additional data. Our data revealed that the analytical method using MSE is useful for unveiling tissue-specific metabolism in vivo.
