Site-specific glycosylation analysis of recombinant human Laminin-511E8 by LC/MS/MS

Laminin-511 E8 is a glycoprotein consisting of α5, β1 and γ1 chains. Recombinant laminin-511 E8 is often used for a culture of stem cells as a matrix. Although this protein has ten potential N-glycosylation sites, neither glycosylation site nor glycan structure is still known. We conducted site-specific glycosylation analysis by proteases digestion, glycopeptide-enrichment, and nanoLC/MS/MS. We demonstrated that most glycosylation sites were occupied with fucosyl biantennary glycans.
