Improvement of 13C Metabolic Flux Analysis by measuring cell components.

The cell composition measurement using GC-MS was applied to budding yeast, a model eukaryote organism. Amount of 14 proteinous amino acids in the four budding yeast strains were successfully quantified by GC-MS. In the literature values, the composition of methionine, alanine, Asx (=asparagine + aspartate), lysine and valine were overestimated as compared with the measured values. BY4947 and RedStar showed high protein ratios in the dry cell weight. Flux distributions calculated using the amino acid composition of literature values and the measured amino acids were compared.
