Effect of ammonia eluent on detection sensitivity in ESI-MS

We compared the effects of ammonia, ammonium acetate, formic acid and ultrapure water on the elution of fat-soluble vitamins and basic compounds in FIA/MS. We discovered that tocopherols were strongly detected only under the ammonia eluent condition. The ratio of signal to noise of the tocopherols were improved by using ammonia eluent in LC-MS/MS. We considered that is due to dissociation of phenol because of the pH of ammonia eluent is 10 and pKa is around 9. Also, the ratio of signal to noise of the diazepam was improved by using the eluent in LC-MS/MS. We inferred that is due to the promotion of ionization or reduce of adsorption to column.
