Absolute Quantification of N-linked Oligosaccharide by Monosaccharaide Analysis

Monosaccharide analysis was applied to isotope-dilution mass spectrometry to accomplish absolute quantification of N-linked oligosaccharide. The hydrolysis condition is examined using six N-linked oligosaccharides and determined to be 110 °C for 4 hours with 2 M trifluoroacetic acid. Under the optimized condition, the measured concentrations of each constituted monosaccharid showed a good agreement each other and the highest result among the measured monosaccharides, which represented complete hydrolysis of the analyte oligosaccharide. This method allows accurate quantification for oligosaccharide.
