メタルフリーカラムを用いたLC/MS/MSによるマウス肝臓中のCoA類のプロファイリングと遊離脂肪酸との相関
Fatty acyl-CoA esters tend to adsorb under the reversed-phase LC conditions. They were eluted as broad peaks and low peak intensity using a stainless steel column. In this study, we aimed to detect sharp peaks of fatty acyl-CoA esters by using a metal-free column, and bio-inert HPLC coupled to tandem MS, and adding ammonium bicarbonate to mobile phase. Twenty-three molecular species of fatty acyl-CoA esters, i.e. CoA to C20:5-CoA, extracted from mouse liver were detected within 15 minutes. The peak area were highly reproducible. The changes of amount of CoAs in mouse liver with fasting were similar to those of free fatty acids.
