Essential metabolic pathways to maintain biological activities include glycolysis, pentose phosphate pathway, tricarboxylic acid cycle, amino acid, and nucleic acid metabolism. Almost all intermediates of these metabolic pathways are sugar phosphates, organic acids, amino acids, and nucleic acids. Therefore, a profiling for these hydrophilic metabolites provides valuable insight towards understanding the different biochemical profiles of a biosystem. In this study, we established a high-accuracy and robust method that enables the comprehensive analysis of polar metabolites by a combination of ion chromatography quadrupole Orbitrap mass spectrometry (IC/Q Exactive), and pentafluorophenyl propyl (PFPP) based liquid chromatography/Q Exactive (PFPP-LC/Q Exactive), and an automated library-based compounds identification. The developed analytical system is a potentially useful tool in the field of life science, medical, and pharmaceutical research.
