HE-ETDにおけるペプチド解離機構解明のためのタンデム飛行時間型質量分析計の開発

High-energy electron transfer dissociation (HE-ETD) is a dissociation process that occurs when the precursor ion collides with an alkali metal gas with keV of kinetic energy. Upon HE-ETD of multiply protonated peptides such as doubly protonated bradykinin formed by ESI, the product ion spectra could be considered as sum spectra of a high-energy collision induced dissociation (HE-CID) parts and an electron capture dissociation (ECD) parts. It is interesting how size and charge state of precursor ion affect to the cross section of collisionally activated processes and that of electron transfer processes. We have developed a tandem time-of-flight mass spectrometer that is suitable for comparison experiment of HE-CID and HE-ETD. As a demonstration, HE-CID and HE-ETD of doubly protonated bradykinin was measured. With high resolving power of the new instrument, it was confirmed that the HE-ETD product ion spectrum of doubly protonated bradykinin could be considered as a sum spectra of HE-CID and electron transfer dissociation.
