アミノ基誘導体化試薬を用いたヒト血漿中微量アミノ酸代謝物の超高感度定量法

We had developed the highly sensitive targeted metabolomics method to analyze the low abundant amino compounds in human plasma. We previously developed a high throughput method for the simultaneous quantification of amino compounds using 3-aminopyridyl-N-succinimidyl carbamate as a derivatization reagent combined with LC-MS/MS. In this paper, we will report the newly developed 19 minutes high throughput LC-MS/MS method to analyze 40 low abundant metabolites having amino group with high accuracy and precision. Thirty-five of these amino compounds in human plasma showed good linearity, limits of quantification, accuracy, precision, and recovery characteristics in plasma by validation studies with scheduled selected reaction monitoring (sSRM) acquisitions. And then, we applied this method for the plasma samples from 10 healthy volunteers. The results revealed that 27 amino compounds, including kynurenine, serotonin, spermine, ADMA and SDMA, were detected in at least one of the 10 samples, and that 24 of these compounds could be quantified. For instance, we could quantified the 31.7 nM for spermine in plasma sample. Moreover, this new method successfully quantified metabolites in plasma sample with high accuracy across over three orders of magnitude, which means to have a potential to cover from low to high metabolite quantitation in the single run.
