Development of 13C metabolic flux analysis for metabolic charaterization of cultured mammalian cells
A method for 13C-based metabolic flux analysis was constructed to characterize metabolic phenotypes of cultured mammalian cells. p53-null soft tissue sarcoma cells were cultured in DMEM containing [1-13C] glucose or [1-13C] glutamine, whose 13C-labeling of intracellular metabolites was measured by GC-MS. A metabolic flux distribution was successfully determined from the 13C-labeling data. The results showed that cancer specific metabolism including high glycolytic and reductive glutamine metabolism was active in the p53-null soft tissue sarcoma cells.
