Disulfide bond formation is one of the most common posttranslational modifications. Many naturally occurring peptides contain multiple disulfide bonds, which stabilize the tertiary structure necessary for activity. Thus, the determination of disulfide bonding patterns is important in studying the biological functions of peptides. Assignment of the pattern is often carried out based on the LC/MS analysis of enzymatic digests of peptides without reduction of disulfide bonds. In this study, we utilized the MS/MS analysis of the digests to investigate the disulfide binding patterns of scorpion toxins.
