Thermal explanation and quantification of MALDI peptide ionization

We found that the spectral pattern of same Tearly were same. The direct proportionality between analyte ion signal and its neutral has been confirmed by calibration curves drawn by [A+H]+/[M+H]+ vs. [A]/[M] not by [A+H]+ only. We also found this linearity is holds when the matrix signal is supressed less than 70% of ion signal of matrix only.
In short, peptide quantification is feasible when spectra are recorded under a constant temperature and low matrix suppression, and the ion signal is expressed in analyte-to-matrix ratio.
