タバコ煙水抽出物との反応により産生される細胞内GSH修飾物質のLC/MS分析
Cigarette smoke extract contains many oxidants and induces injury of various cells. We have studied the identification of active ingredients in water extracts of cigarette smoke (CSE). In a previous study, we determined that glutathione in mouse melanoma cells was modified by methyl vinyl ketone (MVK), which is an α, β-unsaturated carbonyl compound. Glutathione (GSH) is a major detoxication agent in cells. In this study, to clarify the time required for the reaction, we exposed mouse melanoma cells to CSE and used LC/MS to measure changes in GSH levels and its modified products in the cells. We found that the concentration of GSH rapidly decreases within 5 minutes after exposure and the concentration of reaction products (<I>m/z<I> 378 and <I>m/z<I> 366) increases. The peak of <I>m/z<I> 366 was estimated as reduction product of GSH-adduct with acrolein (ACR).
