ブレオマイシン誘発肺線維症モデルラットを対象とした脂質メタボローム解析

Pulmonary surfactant is a complex of lipids (90%) and proteins (10%). It has been indicated that the composition of lipid molecular species in pulmonary surfactant is strongly related to the functions of alveoli. However, comprehensive and detailed profiling of individual lipid molecular species in pulmonary surfactant was not performed sufficiently. In this study, we performed lipidome analysis of bronchoalveolar lavage fluid (BALF) and plasma collected from bleomycin-induced pulmonary fibrosis model rats. More than 100 molecular species were detected from rat BALF using a lipidomics platform based on liquid chromatography coupled to Orbitrap mass spectrometry. Changes in the profiles of individual lipid molecular species in BALF and plasma were observed at the stages of acute inflammation and fibrosis. Based on these results, it was demonstrated that lipidome analysis of pulmonary surfactant and plasma was a powerful tool for biomarker discovery and elucidation of mechanisms of lung diseases.
