Comparative N-Glycomics of 2D- and 3D-Cultured Colorectal Cancer Cell Lines

Colorectal cancer (CRC) exhibits epithelial–mesenchymal heterogeneity linked to malignancy. We performed N-glycomics of five CRC cell lines under 2D and 3D cultures using sialic acid linkage-specific alkyl amidation (SALSA). Abundances of complex-type were remarkably increased in 3D-cultured in comparison with 2D-cultured. In contrast, high-mannose type in 3D-cultured were not changed. It was suggested that complex-type N-glycans play an important role in the formation of CRC spheres.
