Development of a Method for Separating, Identifying, and Quantifying BMP and PG Structural Isomers Using C18 Column UHPLC-HRMS/MS

In lipidomics, UHPLC-HRMS/MS analysis using a reversed-phase column is commonly used, but when the differences in MS/MS spectra between isomers are unclear, analysis is often abandoned due to concerns about the risk of misidentification and quantification. Phosphatidylglycerol (PG) and bis(monoacylglycero)phosphate (BMP) are lipid classes with high biological quantitative value related to lysosomal and mitochondrial function. However, they are structural isomers with very similar MS/MS spectra, making them difficult to distinguish, and there are very few publications describing or discussing them, both targeted and untargeted. We therefore performed detailed analysis of diacylglycerphospholipids in analytical data using a C18 column and found that the retention times (RT) of BMP, PG, and PA molecular species can be estimated from the RT information of PI molecular species, enabling the separation, identification, and quantification of BMP and PG.
