Highly Sensitive Analytical Methods for Cyclic Oligosaccharides and Cyclic Oligopeptides using MALDI-MS

Cyclic oligosaccharides and cyclic oligopeptides have become increasingly important in the pharmaceutical field. Here, we report a highly sensitive and preferential analytical method for these cyclic oligomers using matrix-assisted laser desorption/ionization mass spectrometry (MALDI-MS). The [M+Na]+ and [M−H]− of β-cyclodextrin and [M+Na]+ of cyclosporin A were preferentially and sensitively detected by MALDI-MS when 1-phenylethyl alcohol (1-PEA), particularly (S)-(−)-1-PEA, was used as the analyte solvent in combination with CHCA as a matrix. Under these optimized conditions, acyclic peptides were barely ionized, demonstrating a unique feature that enables selective and highly sensitive analysis of cyclic oligomers.
