Characterizing Lipid Nanoparticle Vaccines Using CDMS and FFF-MALS: A Multi-Dimensional Approach

Lipid nanoparticle (LNP) vaccines have revolutionized nucleic acid delivery, yet their structural complexity and polydispersity pose significant challenges for analytical characterization. Conventional techniques often fail to capture the full spectrum of particle attributes, including mass, size, and heterogeneity, which are critical for understanding formulation quality and performance. In this study, we present a multi-dimensional approach combining Charge Detection Mass Spectrometry (CDMS) with Field-Flow Fractionation coupled to Multi-Angle Light Scattering (FFF-MALS). This orthogonal strategy provides complementary insights into particle mass and size distributions, enabling a more complete characterization of LNP vaccine formulations and supporting the development of robust analytical workflows for next-generation therapeutics.
