Rapid Analysis of Yeast Culture Supernatants Using PESI-MS

Probe electrospray ionization-mass spectrometry (PESI-MS) directly ionizes metabolites adhering to the tip of a probe by applying a high voltage. The method is expected to enable rapid analysis of samples containing large amounts of salts, such as culture supernatants, since the injection of picolitre-level sample volumes minimizes the risk of mass spectrometer contamination. Here, we evaluated a high-throughput method for the analysis of spent medium supernatant from yeasts culture using PESI-MS. Measurement stability and analysis time of amino acids, organic acids and sugar alcohols were investigated. The levels of metabolites in 126 yeast culture supernatants (14 yeast strains × 3 culture conditions × biological triplicates) were measured at 15 seconds per sample, demonstrating the utility of PESI-MS for the analysis of samples containing salts.
