Verification of Matrix and Fragmentation in GAG Structural Analysis Using MALDI-TOF MS

Glycosaminoglycans (GAGs) are widely distributed polysaccharides composed of repeating disaccharide units containing acidic sugars and heterogeneous sulfation patterns. Their structures vary depending on the microenvironment, and disease-specific alterations have been reported in inflammatory disorders. If detailed structural features of GAGs could be used as diagnostic indicators, their application to various diseases would be promising. This study focused on MALDI-TOF MS as a GAG structural analytical method. Previous studies demonstrated that GAG structural analysis by MALDI-TOF MS is feasible with pretreatment procedures, including chemical modification to suppress sulfate loss and complex purification steps. Therefore, we aimed to establish a pretreatment-free method for GAG structural analysis using MALDI-TOF MS by optimizing matrix conditions and investigating the fragmentation behavior of ionized GAGs.
