Visualization of Drug Distribution in the Mouse Brain using t-SPESI-MSI

We developed tapping-mode scanning probe electrospray ionization (t-SPESI) system incorporating tandem mass spectrometry via oxygen attachment dissociation (OAD) and applied it to mass spectrometry imaging of mouse brain sections after administration of drugs (topiramate (TPM), zonisamide (ZNS), and acyclic retinoid phosphatidylcholine (ACR-PC)). The ions of TPM exhibited higher signal intensity in white matter but lower intensity in pyramidal and granular cell layers. The ions of ZNS were detected in negative ion mode measurement with distribution observed throughout the brain, although their intensities were low in the white matter and cerebellar granular cell layer. The ions of ACR-PC were detected in liver but not in brain tissue.
