Evaluation of the Tumor Origin of Serum Biomarker Candidates via Glycoproteomics of FFPE Tissue Sections

Evaluating the tumor origin of biomarker candidates identified by serum glycoproteomics is crucial for ensuring clinical reliability. However, the structural complexity of tumor tissue, including vascular and stromal components, complicates direct attribution of serum-increased glycoproteins to tumor cells based solely on whole-tissue analysis. In this study, colorectal cancer (CRC) was used as a model to assess tumor origin through glycoproteomics of formalin-fixed paraffin-embedded (FFPE) tissue sections. Proteins extracted from FFPE tissues were digested with trypsin, and glycopeptides were enriched using cellulose-based solid-phase extraction prior to LC/MS/MS to obtain the tissue glycoproteome. Identified tissue glycoproteins were compared with glycoproteins showing increased abundance in CRC serum. Candidate glycoproteins common to both datasets were identified, indicating that at least a subset of serum biomarker candidates originated from tumor tissue. This study presents a comparative glycoproteomic approach for evaluating biomarker origin and supports the validity of serum-based cancer biomarker discovery.
