Development of Single-Cell Mass Spectrometry Imaging Using Oxygen Attachment Dissociation-Tandem Mass Spectrometry and Tapping-Mode Scanning Probe Electrospray Ionization

We developed a molecular surface modification method to improve long-term probe stability in tapping-mode scanning probe electrospray ionization (t-SPESI) for single-cell mass spectrometry imaging (MSI). Surface coating with triethoxy(pentafluorophenyl)silane suppressed time-dependent contamination of the probe tip. Coupling t-SPESI with oxygen attachment dissociation (OAD)-MS/MS enabled layer-specific lipid visualization and discrimination of n-7 and n-9 positional isomers of PC 34:1 in mouse cerebellum.
