質量分析法による有機および無機の同時三次元イメージング分析
LA-ICP-MS is an elemental technique for quantitative and imaging analyses on solid samples. Due to its high ionisation efficiency for most elements in the periodic table, LA-ICP-MS has been utilised for the visualisation of metal distribution in biological samples. On the other hand, this characteristic causes LA-ICP-MS to be unable to acquire molecular information. MALDI-MS has been used for complementary molecular imaging with LA-ICP-MS to obtain both molecular and elemental distribution. Analysis was conducted on separate tissue sections, which results in the difficulties for molecular and elemental comparison due to three-dimensional heterogeneity. Hence, an ideal approach is to obtain both information from a single sample for a better comparison. In this study, we developed an analytical technique which utilises laser ablation as the sampling method to simultaneously acquire molecular and elemental distribution in a single sample. The developed technique was also applied for 3D imaging to pinpoint the position of both chemical species in relation to specific 3D structures within the solid sample. For the construction of 3D image and the relation of both chemical species, a software was also developed. This presentation will introduce the principles of the analytical technique, instrumental setup, and software used in this study.
