ウマ尿中のペプチドのスクリーニング検査法における 選択性と検出感度の向上にむけたFAIMS Pro Duoインターフェースの応用

The use of peptide drugs is completely banned at any levels in horseracing and equestrian sports. Owing to the expected low concentrations of peptide drugs and/or their metabolites in complex equine biological matrices, it’s necessary for a doping control laboratory to develop a sensitive and specific test method for the detection of this class of substances. The authors’ laboratory has long been using liquid chromatography/tandem quadrupole mass spectrometry (LC-MS/MS) for the detection of peptides in equine urine and plasma, however, serious matrix influences were commonly observed for the detection of peptides at sub-ppb levels. In this presentation, a highly selective and sensitive screening method for the detection of over 35 peptides at low pg/mL levels in equine urine was developed using ThemoFisher Scientific Vanquish Flex UHPLC coupled to TSQ Quantis Plus equipped with a high-field asymmetric waveform ion mobility spectrometry (FAIMS) interface. Our results showed that the FAIMS Pro Duo interface could significantly reduce the background chemical noise and co-eluting matrix interferences, leading to improved detection selectivity. With this newly-developed method, the limits of detection (LODs) for some of the targets could reach down to less than 2 pg/mL. Optimization of sample extraction method and validation results will be presented.
