LC/MSによる肺腺がん患者血漿由来細胞外小胞を用いたバイオマーカー探索
Extracellular Vesicles (EVs) contain metabolites, such as nucleotides and lipid species, and transport them playing as the intracellular mediators. However, it is hard to detect these metabolites due to the limit of detection. We firstly established the analytical procedure for metabolic profiling of EVs by means of UHPLC-MS/MS and UHPLC-FTMS analysis. Then, we examined to identify the metabolic variation in plasma EVs derived from lung adenocarcinoma patients and healthy volunteers. As results, cysteine and cystine were significantly higher in the EVs from cancer patient, and observed the variation of lipid species, such as triglycerides and phosphatidylcholines consist with polyunsaturated fatty acids. We surmise that the oxidative stress related metabolites in EVs might be correlated with the progression of cancer, and the mechanism should be elucidated in the future study.
