Effect of Probe Oscillation Frequency on Mass Spectrometry Imaging of Mouse Brain Sections by t-SPESI
Mass spectrometry imaging (MSI) is a powerful method for investigating the distribution of various components in tissue samples. Our group developed a tapping mode scanning probe electrospray ionization method to perform MSI. In t-SPESI, an oscillating probe is scanned over the sample, with charged solvent flow through, at the tip, forming a liquid bridge, and picoliters of solvent absorb and ionize sample molecules and deliver them to the mass spectrometer. . In order to improve imaging quality, it is necessary to understand the vibration of the probe. Therefore, we conducted an experiment to investigate the effect of vibration frequency on MSI, and the relationship between the distance between the tip and the sample and the vibration conditions. The results revealed an attractive interaction that exists between probe and sample in positive mode.
