抗体インタクト分析における微量物質の高感度チャージデコンボリューションアルゴリズム
Analysis of intact biological molecules including proteins, carbohydrates, and nucleic acids is one of the most fundamental and ubiquitous applications of mass spectrometry. Sensitivity to low-abundance species, say less than 5% of the most abundant species, however, has always been limited by ionization propensity, ion transmission, background noise, trap capacity, and spectrum interference. Even if the acquired m/z spectra contain signals for low-abundance species, current charge deconvolution algorithms may not reliably map the signals to the neutral mass spectrum, because the signals are insufficiently distinguished from noise. Here we describe and demonstrate improved detection and quantification of low-abundance species in biopharmaceutical samples, including multispecific monoclonal antibodies (mAbs).
