Glucosylceramide (GlcCer) is regulated by glucocerebrosidase (GBA) and essential for maintaining the biological homeostasis. The enzymatical activity of GBA was related with the expression of disease, such as Gaucher. However, galactosylceramide (GalCer), which is the isomeric component of GlcCer must be separated to evaluate the accurate GBA activity. We therefore examined to separate GlcCer and GalCer by means of ion mobility mass spectrometry (IMS) technique using cyclic IMS to develop the analytical method of GlcCer and GalCer in biological samples. As the result, the 50 passes enabled us to have a good separation (Rs = 0.7) of GlcCer (d18:1/18:0) and GalCer (d18:1/18:0) by infusion analysis of standard solutions (1 mg/mL) by cyclic IMS. The optimal condition would be applied for the LC-cyclicIMS-TOF/MS and DESI-MS imaging analyses.
