タンパク質の包括的構造変化検出を目的とした限定消化法の検討

Proteins are biological elements that regulate cellular functions, and their higher-order structures are closely related to their functions. Proteome-scale structural analysis of proteins is essential to elucidate the molecular basis of the ability of cells to change their phenotype in response to external environmental stimuli. One promising method is Limited Proteolysis-coupled Mass Spectrometry (LiP-MS), which combines limited protein digestion with mass spectrometry. However, the protocol is not yet mature enough, and experimental conditions for effective exploration of proteome-scale structural changes need to be investigated. Therefore, in this study, optimization studies of various experimental conditions, including partial digestion, were conducted to comprehensively detect structural changes related to protein function.
