1細胞ダイレクト質量分析による細胞内オルガネラメタボロミクスの開発
There are increasing demands to study the molecular and cellular behavior of a single cell precisely because the current knowledge in life science was obtained using multiple numbers of cells with little information on localization. We developed “Live Single-cell Mass Spectrometry” technique which allows direct analysis of xenobiotics and endogenous compounds in a living cell. The technique is comprised of two steps; 1. capturing of cellular content from a living cell using a metal coated glass microcapillary under video-microscopic observation, and 2. the capillaries are directly subjected to static nanoelectrospray ionization of the sampled cellular content such as cytosol or organelle using mass spectrometry. Currently, to improve the sensitivity and precision of this method, we are studying the development of an automated sampling system, improvement of data variability due to matrix effects, and improvement of mass spectrometry sensitivity by derivatization.
