メタボロミクスデータとフラックスデータによる代謝リワイヤリング解析法の開発

A novel method to identify metabolic regulatory reactions during metabolic rewiring was developed by integrating the data sets of metabolomics and 13C-metabolic flux analysis and employed for the human breast cancer cell line under treatment with paclitaxel, an antimitotic agent. The substrate accumulation against flux decreases of phosphofructokinase (PFK) and pyruvate kinase (PYK) indicated that the fluxes were mainly controlled by enzyme activity. This method was useful for identifying metabolic regulatory reactions, because PFK and PYK, which were known to regulate glycolysis, were identified to be regulated after paclitaxel treatment.
