血清中の誘導体化アミノ酸と短鎖脂肪酸の網羅分析のための前処理法

We developed pretreatment methods for free amino acids and short-chain fatty acids for in comprehensive metabolomic analysis. A large amount of 3-nitrophenylhydrazine was used as a derivatization reagent for them. As a result, the reagent was detected as high ion intensity signal in obtained LC/MS chromatogram. Conventionally, solid-phase extraction was applied for the removal of the derivatization reagent. However, it is difficult to sufficiently recover and detect the various derivatized free amino acids and short-chain fatty acids without separating completely from the derivatization reagent. Therefore, we investigate the method for removal of excess unreacted derivatization reagent in comprehensive metabolomics.
