LC/MS/MSを用いた位置異性体リン脂質の定量分析法の確立

Docosahexaenoic acid (DHA), one of n-3 polyunsaturated fatty acids, abundantly contained in fish. It has been reported that phosphatidylcholine (PC) binding DHA has numerous physiological effects. PC has two fatty acid binding sites, therefore positional isomers exist when different fatty acids are bound (Type A and Type B as shown in Fig. 1). However, there is no precise quantitative method to distinguish the positional isomers of PC. In this study, we synthesized analytical standards for PC positional isomers and established a quantitative method that discriminates positional isomers. Each calibration curve prepared using synthesized standards of PC positional isomers for LC/MS/MS analysis. As the result, the ionization efficiency was different between positional isomers (Type A and Type B). It shows the ionization efficiency differs depending on the binding position of the fatty acid. Elucidating and correcting for differences in ionization efficiency of PC positional isomers will enable accurate quantitative analysis for lipidome in the future.
