The selection of precursor and product ions with high sensitivity and specificity is important for protein quantifications by Multiple Reaction Monitoring (MRM) assay. We optimized peptide selection and the number of product ions detected for each peptide. As a result, two more enzymes for central carbon metabolism in human breast cancer cell were successfully quantified, suggesting that selecting peptides with appropriate weights and analyzing more than 5 product ion per peptide are suitable for MRM assay.
